Loud clicking noise when peddling uphill in larger gears

Hi everyone! I have a problem which I can't seem to figure out.  Recently my rear-gears have been making a VERY loud clicking sound every rotation of the peddle. This only happens when I am in one of the 6 (out of 8) larger gears in the rear, AND I am going uphill and putting a decent amount of force on the peddles. All other times everything is smooth.

So I put my bike upside down, checked to make sure nothing was loose, and tried manually rotating the peddles with my hands in these larger gears.  Nothing, it doesn't make a sound, and everything looks un-bent.

Now, the clicking sound is starting to get VERY loud, loud enough that it feels like something might break. Does anyone have any suggestions/ideas on what I can do from here?

It could be lots of things.

First; is the noise at the frequency of the wheel rotation, or the crank rotation, or neither?

I had a similar noise, it related to the crank rotation, but the root cause was the spokes in the rear wheel.

It is not spoke rub is it? Click a couple of spokes together and see if it the same noise?

A few things to check:
- pedals tight to cranks
- crank arms tight to bottom bracket
- chainrings tight on crank
- cassette tight on freehub
- rear wheel tight in frame
- inspect frame for cracks
